    Salvianolic Acid B, a Potential Chemopreventive Agent, for Head and Neck Squamous Cell Cancer

    Head and neck squamous cell cancer (HNSCC) is one of the top ten cancers in the United States. The survival rate of HNSCC has only marginally improved over the last two decades. In addition, African-American men bear a disproportionate burden of this preventable disease. Therefore, a critical challenge of preventive health approaches is warranted. Salvianolic acid B (Sal-B) isolated from Salvia miltiorrhiza Bge, which is a well-know Chinese medicines has been safely used to treat and prevent aging diseases for thousand of years. Recently, the anticancer properties of Sal-B have received more attention. Sal-B significantly inhibits or delays the growth of HNSCC in both cultured HNSCC cells and HNSCC xenograft animal models. The following anticancer mechanisms have been proposed: the inhibition of COX-2/PGE-2 pathway, the promotion of apoptosis, and the modulation of angiogenesis. In conclusion, Sal-B is a potential HNSCC chemopreventive agent working through antioxidation and anti-inflammation mechanisms

    ChatAnything: Facetime Chat with LLM-Enhanced Personas

    In this technical report, we target generating anthropomorphized personas for LLM-based characters in an online manner, including visual appearance, personality and tones, with only text descriptions. To achieve this, we first leverage the in-context learning capability of LLMs for personality generation by carefully designing a set of system prompts. We then propose two novel concepts: the mixture of voices (MoV) and the mixture of diffusers (MoD) for diverse voice and appearance generation. For MoV, we utilize the text-to-speech (TTS) algorithms with a variety of pre-defined tones and select the most matching one based on the user-provided text description automatically. For MoD, we combine the recent popular text-to-image generation techniques and talking head algorithms to streamline the process of generating talking objects. We termed the whole framework as ChatAnything. With it, users could be able to animate anything with any personas that are anthropomorphic using just a few text inputs. However, we have observed that the anthropomorphic objects produced by current generative models are often undetectable by pre-trained face landmark detectors, leading to failure of the face motion generation, even if these faces possess human-like appearances because those images are nearly seen during the training (e.g., OOD samples). To address this issue, we incorporate pixel-level guidance to infuse human face landmarks during the image generation phase. To benchmark these metrics, we have built an evaluation dataset. Based on it, we verify that the detection rate of the face landmark is significantly increased from 57.0% to 92.5% thus allowing automatic face animation based on generated speech content.     Large Variation of Mercury Isotope Composition During a Single Precipitation Event at Lhasa City, Tibetan Plateau, China

    AbstractThis study examined for the first time the Hg isotope composition in rain samples from a single precipitation event at Lhasa City (China) on the Tibetan Plateau, the “world's third pole”. Large variations of both mass-dependent fractionation (MDF, δ202Hg from -0.80‰ to -0.42‰) and mass-independent fractionation (MIF, Δ199Hg from 0.38‰ to 0.76‰) were observed, with the latter increasing with time. Our results demonstrated that the large variation of Hg isotope ratios likely resulted from mixing of locally emitted Hg and long-term transported Hg, which were characterized by different Hg isotope signatures and mainly leached by below-cloud scavenging and in-cloud scavenging processes, respectively. Our findings demonstrated that Hg isotopes are a powerful tool for investigating the dynamics of precipitation events and emphasized the importance of systematic monitoring studies of the chemical and isotope variability of Hg and other elements during rainfall events

    Bioaccumulation of Hg in rice leaf facilitates selenium bioaccumulation in rice (Oryza sativa L.) leaf in the Wanshan mercury mine

    Mercury (Hg) bioaccumulation in rice poses a health issue for rice consumers. In rice paddies, selenium (Se) can decrease the bioavailability of Hg through forming the less bioavailable Hg selenides (HgSe) in soil. Rice leaves can directly uptake a substantial amount of elemental Hg from the atmosphere, however, whether the bioaccumulation of Hg in rice leaves can affect the bioaccumulation of Se in rice plants is not known. Here, we conducted field and controlled studies to investigate the bioaccumulation of Hg and Se in the rice-soil system. In the field study, we observed a significantly positive correlation between Hg concentrations and BAFs of Se in rice leaves (r2 = 0.60, p < 0.01) collected from the Wanshan Mercury Mine, SW China, suggesting that the bioaccumulation of atmospheric Hg in rice leaves can facilitate the uptake of soil Se, perhaps through the formation of Hg-Se complex in rice leaves. This conclusion was supported by the controlled study, which observed significantly higher concentrations and BAFs of Se in rice leaf at a high atmospheric Hg site at WMM, compared to a low atmospheric Hg site in Guiyang, SW China

    MicroRNA-211 Expression Promotes Colorectal Cancer Cell Growth In Vitro and In Vivo by Targeting Tumor Suppressor CHD5

    Background: Chromodomain-helicase-DNA-binding protein 5 (CHD5) is a newly identified tumor suppressor that is frequently downregulated in a variety of human cancers. Our previous work revealed that the low expression of CHD5 in colorectal cancer is correlated with CHD5 promoter CpG island hypermethylation. In this study, we investigated the effect of microRNA-211 (miR-211)-regulated CHD5 expression on colorectal tumorigenesis. Methodology/Principal Findings: miR-211 was predicted to target CHD5 by TargetScan software analysis. A stably expressing exogenous miR-211 colorectal cancer cell line (HCT-116 miR-211) was generated using lentiviral transduction and used as a model for in vitro and in vivo studies. The expression level of miR-211 in HCT-116 miR-211 cells was upregulated by 16-fold compared to vector control cells (HCT-116 vector). Exogenous miR-211 directly binds to the 39-untranslated region (39-UTR) of CHD5 mRNA, resulting in a 50 % decrease in CHD5 protein level in HCT-116 miR-211 cells. The levels of cell proliferation, tumor growth, and cell migration of HCT-116 miR-211 cells were significantly higher than HCT-116 vector cells under both in vitro and in vivo conditions, as determined using the methods of MTT, colony formation, flow cytometry, scratch assay, and tumor xenografts, respectively. In addition, we found that enforced expression of miR-211 in HCT-116 cells was able to alter p53 pathway-associated regulatory proteins, such as MDM2, Bcl-2, Bcl-xL, and Bax.     Ten-eleven translocation protein 1 modulates medulloblastoma progression

    Background: Medulloblastoma (MB) is the most common malignant pediatric brain tumor that originates in the cerebellum and brainstem. Frequent somatic mutations and deregulated expression of epigenetic regulators in MB highlight the substantial role of epigenetic alterations. 5-hydroxymethylcytosine (5hmC) is a highly abundant cytosine modification in the developing cerebellum and is regulated by ten-eleven translocation (TET) enzymes. Results: We investigate the alterations of 5hmC and TET enzymes in MB and their significance to cerebellar cancer formation. We show total abundance of 5hmC is reduced in MB, but identify significant enrichment of MB-specific 5hmC marks at regulatory regions of genes implicated in stem-like properties and Nanog-binding motifs. While TET1 and TET2 levels are high in MBs, only knockout of Tet1 in the smoothened (SmoA1) mouse model attenuates uncontrolled proliferation, leading to a favorable prognosis. The pharmacological Tet1 inhibition reduces cell viability and platelet-derived growth factor signaling pathway-associated genes. Conclusions: These results together suggest a potential key role of 5hmC and indicate an oncogenic nature for TET1 in MB tumorigenesis, suggesting it as a potential therapeutic target for MBs

    A Blockchain-Based Trust Model for Uploading Illegal Data Identification

    Malicious users can upload illegal data to the blockchain to spread it, resulting in serious threats due to the tamper-proof characteristics of the blockchain. However, the existing methods for uploading illegal data identification cannot select trust nodes and ensure the credibility of the identification results, leading to a decrease in the credibility of the methods. To solve the problem, this paper proposes a blockchain-based trust model for uploading illegal data identification. The trust model mainly has the following two core modules: Reputation-based random selection algorithm (RBRSA) and incentive mechanism. By assigning reputation attributes to nodes, the proposed RBRSA will select nodes according to reputation values. RBRSA favors the nodes with high reputation value to ensure the randomness and credibility of the identification nodes. The incentive mechanism is designed to ensure the credibility of the identification results through the credibility analysis of the model based on game theory and Nash equilibrium. Identification nodes that identify illegal data correctly will obtain incentives. In order to obtain a higher income, the identification nodes must identify illegal data correctly. Credibility analysis and comparative experiments show that the probability of selecting credible nodes by RBRSA is up to 23% higher than the random selection algorithm. The probability of selecting the nodes with a reputation value of 20 by RBRSA is 27% lower than the random selection algorithm; that is, the probability that RBRSA selects untrusted nodes is lower. Therefore, the nodes selected by RBRSA have superior credibility compared with other methods. In terms of the effect of the incentive mechanism, the incentive mechanism can encourage nodes to identify data credibly and improve the credibility of identification results. All in all, the trusted model has higher credibility than other methods

    Mercury concentration and isotopic compositions in lake waters and lake sediment from Canada and China

    Lakes are important ecosystems where mercury (Hg) may undergo extensive methylation and bioaccumulation in the aquatic food web. It is thus crucial to understand the sources of Hg in lake systems. Mercury isotope composition is a powerful tracer of its sources in the environment. However, previous studies on Hg isotope compositions in lake systems mainly focus on the aquatic organism and sediments, whereas lake water that provides the nutrients for the bottom of aquatic food chain has received much less attention. Here we report Hg isotope compositions in surface water and sediments of 8 freshwater lakes in Ontario, Canada. We determined significantly positive Delta199Hg and Delta200Hg values in surface water of all lakes, which are similar to previously reported isotope signature of local atmospheric precipitation. In contrast, the isotope compositions of lake sediments show predominantly negative Delta199Hg and negligible delta200Hg values that are similar to atmospheric elemental Hg(0) and terrestrial soils and plants. We conclude that the source of Hg in lake surface water is likely dominated by direct atmospheric precipitation, while the Hg in lake sediments is primarily derived from watershed inputs of terrestrial particles. The concurrent odd Hg isotope anomalies in surface waters suggest that methylmercury (MeHg) from the watershed wetland, together with Hg(II) derived from the catchment weathering is likely another Hg source to lake waters besides atmospheric deposition. We suggest that the different Hg sources and isotope compositions between lake water and sediments would result in different isotope signatures for MeHg that is produced in water column and in sediments. Therefore, our study not only elucidates the sources of Hg to lake systems, but also has implications in distinguishing between different MeHg sources and in tracing the bioaccumulation of MeHg in aquatic organism
